Regions
A region is a patch of the part with its own plies. The spar example has 11: the whole web (a whole tooling face) and rails and root pads cut out of it by boundaries drawn on a plane and clipped exactly; the web carries 16 plies (2.144 mm), a top rail adds 4, a root pad 2.

Why not give the whole part one stack?
Because the loads aren't the same everywhere. One stack would be too thick in some places, or too thin in others. Too thick costs weight.
